In order to test which side in a diploid x star mating preferentially goes to endpoint we labeled star cells with Mitotracker^® Red (Molecular Probes) and diploid cells with Mitotracker^® Green.  Mitotracker^® Red was used at a concentration of  (0.1 µg /mL) and Mitotracker^® Green (0.5 µg /mL).  The cells were incubated in Mitotracker^® over night. 

 

 

Stock Solution:   50ug / 500uL DMSO = 100ug/mL  Stock

 

mitoGreen  ( to get 0.5 micrograms per mL, use 5 microliters of stock per mL of cells or

50 microliters of stock per 10 mL cells).

 

mitoRed  ( to get 0.1 ug per mL, or 1 microgram in 10 mL,  use 10 microliters stock per mL,  or 100 microliters stock per 10 mL).
